Preparación de ligandos bimetálicos conjugados con biotina como precursores de proteínas con actividad Huisgenasa

Abstract

[ES] Recientemente, el grupo de investigación del Prof. Fernando P. Cossío, en colaboración con los grupos de investigación de los Profs. Cortajarena y Jiménez-Barbero, ha demostrado que las proteínas de repetición tetratricopeptídica consensuada (CTPR) exhiben actividad Huisgenasa en disolución de THF, obteniéndose hasta cuatro cicloaductos racémicos en la reacción entre N-bencilidenglicinato de metilo y (E)-β-nitroestireno. Teniendo en cuenta este precedente, el principal objetivo de este proyecto es continuar con esta línea de investigación mediante la modificación de proteínas para convertirlas en catalizadores de cicloadiciones (3+2), en su versión enantioselectiva. Con este fin, se han preparado distintos ligandos bimetálicos con biotina cuya unión a proteínas debe dar lugar a actividad Huisgenasa.

[EN] Recently, the research group of Prof. Fernando P. Cossío, in collaboration with the research groups of Profs. Cortajarena and Jiménez-Barbero, have shown that consensual tetratricopeptide repeat proteins (CTPR) exhibit Huisgenase activity in THF solution, obtaining up to four racemic cycloadducts in the reaction between methylbenzylidene glycinate and (E)-β-nitrostyrene. Taking this precedent into account, the main objective of this project is to continue this line of research by modifying proteins to convert them into (3 + 2) cycloaddition catalysts, in their enantioselective version. Within this context, different bimetallic ligands with one biotin unit have been synthesized as suitable ligands to be bond to proteins which should exhibit Huisgenase activity.
